GRO (Growth Related Oncogene) Overview

  • Family: GRO belongs to the IL-8 family of chemokines, which are known for their roles in inflammation and immune responses.
  • Isoforms: GRO is known to exist in three isoforms:
    • GROα (MGSA - Melanoma Growth Stimulating Activity): Shares high homology with IL-8.
    • GROβ
    • GROγ

Molecular Characteristics:

  • Identity with CINC-1: Each GRO isoform shares significant sequence identity with CINC-1 (cytokine-induced neutrophil chemoattractant 1), with 68% identity for GROα, 71% for GROβ, and 69% for GROγ. CINC-1 is considered a rat counterpart of human GRO proteins.
  • GROα/MGSA: Shows high amino acid sequence homology with IL-8 and exhibits similar biological activities.

Functions:

  • Inhibition of Endothelial Cell Proliferation: GRO is known to inhibit the proliferation of endothelial cells, which are critical for the formation of new blood vessels.
  • Leukocyte Chemotaxis: GROα exhibits significant chemotactic activity, recruiting leukocytes to sites of inflammation, similar to IL-8.
  • Inhibition of Angiogenesis: Specifically, GROβ has been reported to inhibit angiogenesis when administered in vivo. This indicates its potential role in controlling blood vessel formation, which is important in various physiological and pathological conditions.

Production:

  • Source: GRO proteins are produced by various cell types, including:
    • Activated Monocytes
    • Fibroblasts
    • Epithelial Cells
    • Endothelial Cells

Functional Implications:

  • CINC-1 Relation: GRO proteins, including GROα, GROβ, and GROγ, were initially purified from media conditioned by IL-1β-stimulated rat kidney epithelial cells (NRK-52E). This suggests a functional similarity between CINCs and human GROs.
  • Anti-Angiogenic Role: The ability of GROβ to inhibit angiogenesis has implications for cancer research and other conditions where uncontrolled blood vessel growth is a problem.
