Gastric Inhibitory Peptide (GIP) Overview

Function and Role:

  • Incretin Hormone: GIP is a gastrointestinal hormone that stimulates insulin secretion from pancreatic beta cells in response to food intake and inhibits glucagon release from pancreatic alpha cells.
  • Nutrient Regulation: Plays a key role in regulating nutrient retention and metabolic processes in various cell types, including pancreatic beta cells, adipocytes, and osteoblastic cells.

Biochemical Characteristics:

  • Discovery: GIP, initially known as Gastric Inhibitory Peptide, was first identified and sequenced from intestinal mucosa in 1970. It was later renamed glucose-dependent insulinotropic peptide.
  • Forms:
    • Active Form (GIP 1-42): The biologically active form of the peptide.
    • Inactive Form (GIP 3-42): Formed through rapid inactivation of GIP 1-42 by the enzyme DPP-IV (Dipeptidyl Peptidase-IV).

Receptor and Expression:

  • Receptor Presence: The GIP receptor is expressed in several cell types:
    • Pancreatic Beta Cells: Where it stimulates insulin secretion.
    • Adipocytes: Involved in fat metabolism and energy regulation.
    • Osteoblastic Cells: Affecting bone metabolism and growth.

Clinical Relevance:

  • Metabolic Syndrome: Modulation of GIP signaling is considered beneficial for improving conditions related to metabolic syndrome, such as type 2 diabetes and obesity.
