Amyloid-beta (1-42) ELISA Overview

Purpose: This ELISA assay is designed for the measurement of Amyloid-beta (Aβ) peptides, specifically Aβ (1-42), which are cleaved at the N-terminal side. The assay is particularly useful for analyzing Aβ variants that play a role in Alzheimer’s disease.

Background:

  • Amyloid-beta (Aβ): Aβ peptides are derived from the amyloid precursor protein (APP) through the actions of β-secretase and γ-secretase. These peptides are implicated in the formation of amyloid plaques, which are a hallmark of Alzheimer’s disease.
  • Variants: The Aβ peptide can be 40 or 42 amino acids long, with Aβ (1-42) being one of the more prominent forms found in Alzheimer’s plaques.

Significance:

  • Alzheimer’s Disease: The accumulation of Aβ plaques in the brain is a key pathological feature of Alzheimer’s disease. The Aβ (1-42) form is particularly important due to its propensity to aggregate and form plaques.
  • Discovery: In 1995, research by Saido et al. highlighted the differential deposition of amyloid-β peptide species, including Aβ (N3pE). This variant, starting at the 3rd amino terminal residue glutamate, converts to pyroglutamate through intramolecular dehydration, a modification linked to the progression of Alzheimer's disease.
