Peripheral Blood Mononuclear Cells (PBMCs) form part of white blood cells, with a round nucleus and consists of lymphocytes (T cells, B cells, NK cells), monocytes and dendritic cells.

A primary step and the most common PBMC isolation method involves using a density gradient medium and centrifugation. Whole blood is first diluted with phosphate buffered saline (PBS) and then carefully layered over the density gradient medium.
